Amylum diambil dari farinanya yaitu bentuk pati kasar ditambah air, kemudian disaring. Filtrat yang didapat kemudian diendapkan. Endapan tersebut diambil sebagai amylum setelah dikeringkan.
Yang harus diperhatikan pada pengamatan amylum yaitu : bentuk hilus, dan lamela, serta ukurannya. Secara organoleptis warnanya putih, bentuk padatan, tidak berbau, bila ditambah aqua – Iod akan berwarna biru.
- Amylum oryzae (Pati Beras)
Tanaman : Oryza sativa L
Familia : Poaceae/graminae
Simplisia : Terdiri dari butir – butir tunggal/ majemuk.
Bentuknya segi banyak (polygon)
Ukurannya 5 µm
Umumnya tidak mempunyai hilus, kadang – kadang terdapat hilus pada pembesaran kuat.
- Amylum Manihot (Pati Singkong)
Tanaman : Manihot utilissima
Familia : Euphorbiaceae
Simplisia : Terdiri dari butir – butir tunggal/majemuk
Bentuknya bulat, kadang – kadang ada bentuk seperti helm (topi baja)
Hilusnya ditengah, lamela tidak jelas. (kecuali pembesaran kuat). Ukurannya 20 µm
- Amylum Solani (Pati Kentang)
Tanaman : Solanum tuberosum
Familia : Solanaceae
Simplisia : Terdiri dari butir – butir tunggal
Bentuknya oval, ukuran 100 µm
Hilus eksentris (dipinggir), lamela jelas
- Amylum Tritici (Pati Terigu/gandum)
Tanaman : Triticum vulgare
Familia : Graminae
Simplisia : Terdiri dari butir – butir tunggal/majemuk
Bentuknya bulat, ada yang besar 30 – 40 µm, ada yang kecil 10 – 20 µm.
Hilus dan lamela tidak jelas.
- Amylum Maydis (Pati Jagung)
Tanaman : Zea mays
Familia : Graminae
Simplisia : Terdiri dari butir – butir tunggal/majemuk
Bentuknya polygonal, ukuran 5 – 25 µm
Hilus konsentris (ditengah) dan retak, lamela sulit dilihat.
